CD8mAb对呼吸道合胞病毒诱导的哮喘小鼠气道炎症的影响
【关键词】 呼吸道合胞病毒；哮喘；CD8 T细胞；CD8单克隆抗体；气道炎症【Abstract】 AIM: To evaluate the effect of antiCD8 monoclonal antibody (antiCD8mAb) on airway inflammation of Balb/c mice with respiratory syncytial virus(RSV)induced asthma. METHODS: Fifty mice were randomly divided into 5 groups (n=10 each group): PBS control group, OVA group, OVA/RSV group, antiCD8mAb treated group and IgG2αmAb treated control group. Lung histopathological changes were evaluated by HE stain. Airway responsiveness was assessed using a whole body plethysmography. Cells in bronchoalveolar lavage fluid(BALF) were counted and classified and the supernatants of the BALF were used for detection of interleukin（IL）4, IL5 and interferon（IFN）γ. The percentage of CD4 （IFNγ , IL4 , IL5 ）, CD8 （IFNγ , IL4 , IL5 ）T lymphocytes and the ratio of Th2/Th1, Tc2/Tc1 in peribronchial lymph nodes(PBLN) were analysed by threecolor flow cytometry method. RESULTS: Intraperitoneal administration of antiCD8mAb can remarkably inhibit airway inflammation and airway hyperresponsiveness(AHR).
